I used one code to extract tumour from the images. I am able to extract them when the tumour background is black. i.e, for untitled12,untilted123 images. Im not able to extract the tumour when the tumour is black. Ive tried invert colours but im not getting the output. Image is Slice104.

Threshold for dark stuff rather than bright stuff. Just use
grayImage < someThreshold;

Like I said, you have to write the code that determines the threshold from the histogram. There is no one algorithm that works in all cases.
I'm attaching an example tumor finding program.
