I am trying to segment an X-ray scan of the knee joint, to separate the bone from tissue and ligaments.

See my File Exchange for an image segmentation tutorial. However it doesn't work at finding every single thing you could possibly imagine in every possible image ever created in the universe over all time. As you can imagine, an algorithm that could pick out a plane against a clear blue sky won't be the same algorithm as one that would pick out male persons from a crowd at a stadium, or certain biological organs from biological images. So that means that my demo probably won't precisely pick out whatever object you're interested in in your picture. Obviously you'll have to take general image processing concepts and apply them in the correct order for your specific situation.

same goes for me i'm actually a begginer first time working with matlab and i need to create an algorithme for seggmentation and thresholding the lunate from the carpal bones so i can perform 3D reconstruction and create a 3D module similar to the original bone and then print a 3D prosthesis model, i really need help

@manal hamda, I haven't seen your image but if it's like a lot of radiographs, the object of interest is not well separated and thus will be very hard to isolate from other, surrounding tissues. This is probably too challenging of a project for a beginner. I suggest you go here for professional help, if you really need it done :
