Can anyone help me with calculating the 1st derivative of a ppg signal ?

I tried using the diff command but later i realized that its just taking the difference.So can anyone suggest me how to go about calculating the 1st derivative of the ppg signal.

Are the associated time intervals all equal or are they unequal? It makes a difference in the complexity of the derivative expression.
